Will doing this damage the caliber or the mount, the fork or the wheel?
Are disc brakes designed to work backwards?

I can't imagine that you can go backwards with enough force to damage the mount on the frame or fork, nor the caliper, rotor, or wheel. The last three don't 'care' what direction things are going, and the first two have probably been over-engineered for safety on any consumer available product.
