Hamstrings Twitching (mostly one)

I am a 41 yo male triathlete and have been increasing my cycling distance over the past 6-9 months. In June I purchased a new TT bike (cervelo p3) after having ridden an old 650cc QR Kilo for the past 2 years (going far fewer miles then). This summer I wanted to try a longer triathlon (the bike leg in the past never went over about 25 miles), so I increased my distance slowly up to the 56 miles necessary for the 1/2 IM this summer. I had no problems at all over the summer with my legs, but as a triathlete I usually only ride 3 times a week about 4 hours total, though I did a few weeks of 6-7 hours. When I rode my long ride (56-60 miles) I got a numb perineum that would not go away until I came out of aero position for at least 10 minutes. This only happened 2-3 times.

This fall I bought a computrainer for winter training (I live in a snowy area). I have been riding that (same bike) about 3-4 hours a week and began to get the numb perineum again; now with every ride and earlier into it. I do not like a numb perineum, so after a bit of research I bought a new Adamo TT saddle (it has no nose). It has been about 6 weeks of riding. The perineal problem has been completely gone since ride one. I have gone up to about 2.5 hours on it without a problem, except that my 'sit' bones are sore (I expected that) and I was rocking back and forth a bit on the bike, which I read meant I was a little too high. The saddle instructions mention that the seat post usually has to be lowered for proper positioning. I originally had a professional bike fit with my purchase, but live too far from the shop for that to be convenient to do again. I have been lowering my seat post by milimeters each ride trying to find the spot where I don't rock.

Now I will get to the point of my post! 2 days ago I did a fairly standard 1 hour ride and for the past 3 days my hamstrings (mostly just my right medial one) have been twitching. It is annoying, not painful. It comes, lasts about 5 minutes and goes away. Does this have to do with my seat post? Can I ride again today or will I make it worse? Will it go away in time without causing any real damage, or will it turn to a painful cramp?

I have had twiching before usually in my calves after tough workouts (sometimes after a run) and that went away when I started taking extra magnesium (I take zegerid, which can leech away magnesium) and the twitches have been gone 6 months until now. I have been taking magnesium for the past few days like its my job, but to no avail. I should mention that the day after the ride I went for a 6 mile run (not long at all for me), it was cold outside (32F, 0C) but I was bundled for it. My quads were suprisingly sore after that run (unless that was from the ride?).

Anyway, sorry for the long post, but I wanted to give all the details. I appreciate any help I can get.
